The closest commercial airport is Flagstaff Pulliam Airport which is about a 45 minute drive from Williams. The nearest International airport is Phoenix Sky Harbor air port (PHX) that is a 2 1/2 hour trip give or take traffic and weather. Las Vegas is another option and can sometimes be a cheaper option if you don’t mind the 3 hour drive. Amtrak does make some late night stops at Williams Junction, then a bus will bring you into town or else you can use the Flagstaff Amtrak depot. There is also Greyhound.
There are 3 exits from I-40 into Williams. Exit 163 is the main exit/entrance into the heart of downtown Williams. If you want to just get to the Grand Canyon take exit 165 which takes you to Rt 64. That takes 1 hour (60 miles) roughly to get to the canyon.
